"Did you just assume my gender" is a common joke by reactionaries mocking transgender people for having preferred pronouns, even though no one ever actually says this in real life. It's a derogatory stereotype.

imo it isn't immediately clear what is offensive about the tweet based just on what is in the OP.

Even if you don't know why it's transphobic just ask "why is it transphobic?" so you can learn instead of saying "put down the pitchforks". Jesus it's not that hard to ask and learn so you can understand why it's offensive.
The OP is just a single line tweet, for people that weren't familiar with the background of this phrase it is confusing what the transphobic nature of it is. So it sort of makes sense to ask for context as to why it is offensive?
